高等細胞の免疫系は、T細胞、B細胞,樹状細胞(dendritic cell, DC)等の免疫細胞により、抗原の提示、細胞表層リセプターを介した細胞間のコミニュケイション等により巧妙に制御されています。当研究室では、免疫制御物質ならびに 細胞表層リセプター等の詳細な研究を行い、そのリコンビナントプロテイン或いは改良型プロテインを作成し、それにより免疫系を正、負両方向にコントロールし,癌ならびに自己免疫疾患等の治療に貢献する事を目的としております。3年程前、英国のオックスフォード大学で制御性T細胞(Treg)、DCの活性を制御するサイトカイン、TNFならびにTNFリセプターファミリー(GITR, GITR ligand, CD40等)の研究を行っていた研究グループを新たに加え Treg細胞の分化、それらの遺伝子発現調節機構の解明へとその研究の幅を広げ、更にT細胞ならびに抗原提示細胞(DC等)を介した免疫系の活性化機構の解明、治療への応用へと発展させる事を考えております。

今回、こちらの研究者の帰国、移動に伴いポスドクのポジションに空きが出来ます。研究には、癌ならびに自己免疫疾患のモデルマウスを使用する事もありますので、マウスを用いた研究の経験者は歓迎されますが、やる気のある方であればこちらで技術を習得する事も可能です。

研究室はハーバード大学Korsmeyer研究室から独立したDr.James Hsieh(Taspase-1/MLLグループ)とDr.Emily Cheng(アポトーシスグループ)の夫妻で共同運営されています。Hsiehグループでは遺伝子改変マウスから培養細胞まで様々なレベルでTaspase1及びMLL1/2の機能解析を行っています。基本的には一人1-2テーマを独立に担当します。セミナーなどは2グループ合同で行っており、実験室・居室も共通で使用しています。2人のPIはともにRO1他複数のグラントでサポートされており、研究機器なども充実していています。研究室構成は現在PI2名、テクニシャン2名、ポスドク3名(日本人1名)、学生4名(日本人1名)です。

NIAID/NIH Postdoc募集

Postdoctoral Fellow Position in Structural Biology of Innate Immune Response

A postdoctoral fellow position is immediately available in the Structural Immunobiology section, Laboratory of Immunology (NIAID/NIH) for a highly motivated individual with a strong interest in signaling mechanisms of the innate immune responses. The innate immune system serves as the gatekeeper and coordinator for the defense against microbe infections. One of the paradox of the innate immune receptors is that they are specific for each class of pathogen antigens, whereas not too stringent to render them non-responsive to similar but different epitopes. How do the receptors achieve such “broad” specificity is among the most important questions structural biologists could strive to answer. The projects aim to elucidate how the pattern recognition receptors (PRRs) sense pathogenic epitopes and how this recognition initiates downstream signaling cascades to direct appropriate responses against various pathogens. One of the best approaches to decipher the signalin!
g network is through studying protein-protein interactions involved in the crosstalk, using X-ray crystallography integrated into other biophysical and biochemical techniques. Elucidating the signaling mechanisms in innate immune response has important implications in the prevention and treatment of infectious diseases, autoimmune diseases and cancer.

The lab will be primarily structure oriented, complemented with functional studies. The section is fully funded and equipped with state-of-the-art protein expression, purification and crystallization instruments. There is regularly scheduled beam time slot for NIH at the APS synchrotron for data collection. There are also numerous opportunities for collaboration and acquiring new skills among the 27 Institutes and Centers at NIH with one of the most diverse groups of international researchers.

The ideal candidate must have a Ph.D. in biochemistry/biophysics/X-ray crystallography or a related field, and extensive experience in molecular biology and protein expression/purification. Knowledge in crystallization, data collection, phasing, model building and refinement, and structure analysis is desirable but not required. The candidate must possess excellent writing and oral communication skills, and will be expected to work in a diverse and collaborative environment. Salary will be highly competitive and commensurate with experience and in the $40,000 range, supplemented with excellent fringe benefits.

The laboratory of Dr. Fawaz George Haj at the Department of Nutrition, University of California, Davis is seeking outstanding candidates for a postdoctoral position. Experience in molecular biology, mouse models of obesity/diabetes and/or cell culture is highly desirable.
